Mechanism
CRISPR interference (CRISPRi) enables targeted transcriptional silencing in mammalian cells. A catalytically inactive Cas9 (dCas9) fused to a repressor, such as KRAB, is guided by a customizable single guide RNA (sgRNA) to complementary DNA. The dCas9-sgRNA complex sterically blocks RNA polymerase elongation, effectively repressing the target gene.
