Highlights:

ALSTEM offers a human iPS cell line with inducible expression of Ngn2 by doxycycline (Dox) (iP11NK). This cell line is generated from pre-made footprint-free human iPS cells that have the inducible Ngn2 gene stably integrated into the human AAVS1 “Safe Harbor” site. It provides a convenient means to differentiate human iPS cells to excitatory neurons rapidly and efficiently. The integration at “Safe Harbor” sites ensures safe and reliable expression of Ngn2 when Dox is supplied. Alstem further engineered this cell line by inserting dCas9-KRAB cassette into the cell by lentivirus transduction.  This cell line allows one to suppress the expression of any gene by introducing a gene-specific gRNA that can be cloned into one of Alstem’s gRNA Lentivirus Expression Vectors (a link here to the list of these vectors).

Description:

Neurogenin (Ngn) 2 is a basic helix-loop-helix (bHLHtranscription factor that promotes direct differentiation of pluripotent stem cells to functional excitatory neurons. AAVS1 (also known as PPP1R12C) locus supports robust and sustained transgene expression. Precise insertion of tetracycline inducible Ngn2 into human iPS cells at AAVS1 site could generate a stable iPSC line for rapid neuronal differentiation upon induction.

The ALSTEM scientists developed a highly responsive doxycycline inducible Ngn2-expressing human iPS cell line, which is ideal for in vitro rapid neuronal differentiation. This cell line (Cat No. iP11NK) is generated from pre-made footprint-free human iPS cells by inserting Ngn2 and M2rtTA at the AAVS1 locus by gene editing using the HDR pathway. In the absence of doxycycline (Dox), the cells retain the pluripotent state; however, upon induction by Dox, the cells will differentiate into functional excitatory neurons rapidly in one week.

This cell line also stably expresses a dCas9-KRAB cassette and can be used to inhibit endogenous gene expression when a gene-specific gRNA is supplied. We have validated this gene suppression using CD59-specific gRNA cloned into one of our gRNA Lentivirus Expression vectors.

Quality Control

iNgn2-dCas9-KRAB Human iPS Cells were grown under feeder-free conditions with mTeSR1 medium. Each lot of hiPSCs is tested for growth and viability following recovery from cryopreservation. In addition, each lot is tested for the expression of TRA-1-60 and OCT4.
